Cancer drug names and their functions, mechanisms and side effects

5-fluorouracil
Cancer drug - breast, colorectal, panrceatic, stomach cancers. Side effects: nausea,
diarrhoea, vomiting. Blocks etimodate synthesis.
Cisplatin
crosslinking platinum analogue, interferes with DNA replication killing the fastest
proliferating cells. Side effects: nephrotoxicity
Cyclophosphamide
alkylating crosslinker, high doses: increased cytotoxicity & immunosuppression,
lowdoses: antiangiogenic, immunostimulation. SE: nausea
Dexamethasone
glucocorticoid, steroid, SE: acne, depression, vomiting
Imatinib (Gleevec)
inhibits BCR-ABL, used for CML and GIST (gastrointestinal stromal tumours), SE:
nausea,diarrhoea
Interferon-alpha
angiogenesis inhibitor, SE: flu-like syndrome (within 1-2hrs), fatigue, low blood count
Methotrexate
inhibits dihydrofolate reductase immunosuppressant, SE: nausea, tiredness, low white
cell counts, breakdown of skin in mouth, TERATOGENIC
Paclitaxel (Taxol)
targets tubulin, enhancing spindle formation -- prevents chromosomes form reaching
metaphase -- blocks mitosis progression
Prednisone
glucocorticoid, immunosuppressant, SE: high blood glucose levels, fluid retention,
depression, anxiety // Cushing's syndrome, osteoporosis
Rituximab
B cell depletor: antibody or fusion proteins that bind to CD20 on B-cells, increasing death by
NK cells. SE: cardiac arrest, infections
Tamoxifen
pro-drug, anti-oestrogen, acts as an ER antagonist inhibiting transcription of
oestrogen-responsive genes in tissues, blocks ErbB2/HER2.
Thalidomide
angiogenesis inhibitor, inhibits production of TNF-alpha, IL6, IL10 and IL12, enhances
production of IL2, IL4 and IL5. increases lymphocytes
Trastuzumab (Herceptin)
monoclonal antibody, inhibits HER2, suppresses angiogenesis, SE: flu-like symptoms
Vincristine (Leurocristine / Oncovin)
mitotic spindle poison, binds to tubulin protein, stops the cell from separating
chromosomes in metaphase. SE: chemo-induced p.neuropathy
